To specify an exact XYZ starting location and ending location, use the Extract a Precise Line (Data>Extract Precise Line). If you are running Tecplot 360 in batch mode, you need to make sure that you do not change the orientation of your plot that it stays the same relative to your Polyline. I will turn off the mesh so you will see only the Polyline. One warning about Extracting from Polyline. And if you have transient data, when you have the Polyline selected, you can go to data, and extract the geometry over time. A tool like this is useful to follow the path of that river.
